Went to start up the bike and go for a ride and when it started up I instantly noticed that it was only running on one cylinder. The front cylinder exhaust pipe was cool to the touch and the rear was warm. I pulled the front plug and it looks fine started the bike with the plug grounded out and it had spark. I didn't have fitting for the buell head to check the cylinder compression. I did put my finger over the plug port and it did blow my finger off.(I know thats really backyard, but I am going to go buy an adapter for my compression gauge so I can really check the actual compression. I pulled the fuel rail and both injectors are squirting fuel. I pulled the intake manifold off and the header. The valves are moving. Any suggestions?

Compression, air, fuel, spark....that's really all it takes.

Does it appear that you have a strong spark? I would replace the plugs as a cheap first step. Perhaps you have a bad coil or wire?

Did the plug smell like fuel when you pulled it?

might look at the connector on your front injector. they can come off. No injector no fuel no bang no matter how much spark you have.
Pull the right side air scoop off and look up on the inside of the frame. That is where the injectors plug in. It pushes in at a 45 degree or so angle and has side clips that should hold it in place. This connector is not strong and can loosen if you are unlucky.

Mine did the same thing, changed the plugs and all was fine.

I would try that first for sure.
